Self‐Delivering Microstructured Iota Carrageenan Spray Inhibits Fibrosis at Multiple Length Scales

Fibrotic diseases account for 45% of all deaths in the developed world, and progressive scarring conditions, such as dystrophic epidermolysis bullosa (EB), result a significant reduction quality life. There is thus need novel approaches fibrosis prevention. Herein, it shown that iota carrageenan, low‐cost, regulatory approved polysaccharide, effective at preventing by inhibiting collagen fibrillogenesis, abrogating transforming growth factor beta 1 (TGFβ1) pathway, lubricating epithelium. It demonstrated other anionic polysaccharides, can prevent fibril formation, key step scar formation. also binds TGFβ1 to myofibroblast transdifferentiation, evidenced both transcription translation alpha smooth muscle actin. Iota carrageenan may be formulated microparticle suspension, allow spraying even coverage on hard‐to‐reach anatomical sites, like oral mucosa. This formulation provides good lubrication, reduce shear forces initiate EB. study not only therapy prevention, but demonstrates potential self‐delivering immunomodulatory polysaccharides safe, cost‐effective, stable platform, which more easily translated clinical benefit.
